Canine mitogen-activated protein kinase kinase 4 (MAPK4), also known as ERK4, p63MAPK, and PRKM4, is a non-classical member of the MAPK family that plays a significant role in cellular processes such as proliferation, differentiation, apoptosis, and stress response. Overexpressed in various malignancies, MAPK4 orchestrates cell proliferation, migration, and apoptosis via the AKT/mTOR and/or PDK1 signaling pathways, thereby facilitating tumor cell growth. Its expression is closely associated with the effectiveness of specific inhibitors like PI3K and PARP1, and it correlates with the survival rates of cancer patients. Targeting MAPK4 may offer a new approach to cancer therapies and it has the potential to serve as a prognostic marker in various cancers.
