Porcine phosphoenolpyruvate carboxykinase 1 (PEPCK1) is a cytosolic enzyme that plays a crucial role in gluconeogenesis, the metabolic pathway responsible for generating glucose from non-carbohydrate sources. This enzyme is considered the first rate-limiting step in gluconeogenesis, catalyzing the conversion of oxaloacetate (OAA) and GTP into phosphoenolpyruvate (PEP), CO2, and GDP. In pigs, PEPCK1 is not only pivotal in liver and kidney gluconeogenesis but also has been found to be important in adipose tissue, where it contributes to glyceroneogenesis, a process essential for triglyceride synthesis. Recent studies have shown that overexpression of the cytosolic form of PEPCK in skeletal muscle can enhance intramuscular fat content, which is a desirable trait for meat quality in pigs. This finding suggests that PEPCK1 could be a potential target for genetic modification to improve pork quality.
