Target Information

DNA topoisomerase II alpha (Topo II Alpha) is an essential nuclear enzyme that regulates DNA supercoiling by catalyzing the relaxation of both positively and negatively supercoiled DNA. It plays a critical role in various DNA metabolic processes, including replication, transcription, and chromosome condensation and segregation. Topo II alpha does this by making a transient double-strand break in the DNA, allowing the DNA strands to pass through and rejoin, thus altering the DNA's topological state without altering the base sequence. Topo II alpha is a target for several chemotherapeutic agents, particularly those used in the treatment of cancer. Drugs like etoposide and doxorubicin poison the enzyme, leading to DNA damage and preventing the enzyme from rejoining the DNA strands, which results in cell cycle arrest and apoptosis.
