Canine pseudouridine synthase 7 (PUS7) is a key enzyme responsible for the conversion of uridine to pseudouridine in RNA molecules, playing a critical role in RNA stability and function. This modification, known as the "fifth nucleotide," is abundant in various RNA species and can impact mRNA processing. PUS7 is unique among eukaryotic Pus proteins as it modifies a wider array of substrates and has limited homology with other Pus family members. It recognizes a consensus sequence, UNUAR (where R stands for purine), and is influenced by RNA structure, preferentially modifying less-structured contexts. PUS7's activity is not only confined to non-coding RNAs but also extends to protein-coding mRNAs, with its modification sites varying depending on cellular conditions.
