Colipase serves as a crucial cofactor for pancreatic lipase, facilitating the enzyme's attachment to the lipid-water interface. Produced by the pancreas in its inactive form, known as procolipase, it is activated in the intestinal lumen by the enzyme trypsin. As a bridging molecule, colipase binds to both lipase and bile acids, ensuring that lipase remains anchored to the lipid droplets and is not displaced.
