Canine PAICS (phosphoribosylaminoimidazole carboxylase/phosphoribosylaminoimidazole succinocarboxamide synthetase) is a bifunctional enzyme that plays a crucial role in the de novo purine synthesis pathway, which is essential for the production of nucleotides required for DNA synthesis and cellular proliferation. Overexpression of PAICS has been observed in various cancers, including colorectal, breast, lung, glioma, prostate, and gastric cancer, and is associated with poor prognosis. In dogs, PAICS may serve a similar function, regulating metabolism, cell cycle progression, proliferation, epithelial-mesenchymal transition (EMT), migration, and invasion in cancer cells. Targeting PAICS could be a promising strategy in anti-tumor therapy, as it not only enhances the de novo synthesis of purine nucleotides but also promotes tumor growth and metastasis.
