Porcine glutathione peroxidase 4 (GPX4) is a vital selenoenzyme that plays a critical role in antioxidant defense mechanisms within cells. It is best known for its ability to reduce lipid hydroperoxides to their corresponding alcohols, thereby preventing the accumulation of toxic lipid peroxides that can lead to cellular damage and death. GPX4 is also recognized as the primary regulator of ferroptosis, an iron-dependent, non-apoptotic form of cell death that is mediated by the accumulation of lipid peroxides. In pigs, GPX4's function is particularly important in maintaining the integrity of cellular membranes and in protecting cells from oxidative stress. It has been shown to be involved in the development and survival of photoreceptors in the eye , and its expression is associated with the inhibition of diseases such as neurodegenerative disorders and certain types of cancer. The enzyme's role in regulating ferroptosis makes it a potential therapeutic target for conditions where the balance of lipid peroxidation and antioxidant defense is disrupted, such as in sepsis, ischemia-reperfusion injury, cardiovascular diseases, and cancer.
