Bovine Mothers Against Decapentaplegic Homolog 5 (SMAD5), also known as MADH5 or JV5-1, is a transcriptional regulator activated by BMP type I receptor kinases. It plays a critical role in the BMP signaling pathway, which is involved in embryonic development, cell differentiation, angiogenesis, and tissue homeostasis. SMAD5 is phosphorylated upon BMP ligand binding to its receptors, forming a complex with SMAD4 that translocates to the nucleus and acts as a transcription factor. This protein is also involved in energy metabolism regulation by promoting mitochondrial respiration and glycolysis in response to cytoplasmic pH changes.
