Canine dihydrofolate reductase (DHFR) is a ubiquitous enzyme that plays a critical role in the biosynthesis of tetrahydrofolate cofactors, essential for the transfer of C1 carbon moieties in various biosynthetic pathways, including the synthesis of purines, amino acids like glycine and methionine, and thymidine. This enzyme was discovered as the target for antifolate drugs such as methotrexate in mammalian systems, and also for antibacterial agents like trimethoprim and antiprotozoal agents like pyrimethamine. DHFR's key function in folate metabolism makes it a significant target for drugs used in cancer treatment, antibacterial therapy, and other antiparasitic medications.
