Canine lem domain containing 3 (LEMD3) is an integral inner nuclear membrane protein that antagonizes TGFβ-SMAD2/3 signaling in a stiffness-dependent manner, both in the nucleus and cytosol. It plays a role in the regulation of extracellular matrix (ECM) stiffness and TGFβ activation, which are key drivers of pathological fibrosis in multiple organ systems. LEMD3 binds and inactivates the downstream transcription factor partners of TGFβ and BMP, the receptor proteins' mothers against decapentaplegic (Smads) 2 and 3. This protein's interaction with Smads and its role in mechanotransduction make it a significant target for understanding and potentially treating fibrosis and other ECM-related pathologies in dogs.
