Canine eukaryotic initiation factor 2 alpha subunit (EIF2S1) is a component of the EIF2 complex, which is essential for the initiation of protein synthesis in eukaryotic cells. It facilitates the placement of the initiator tRNA onto the 40S ribosomal subunit during translation initiation. EIF2S1 is regulated by phosphorylation, which occurs on serine 51 and is mediated by various kinases in response to cellular stresses such as heme deficiency, oxidative stress, viral infection, endoplasmic reticulum (ER) stress, and amino acid deficiency. Phosphorylation of EIF2S1 plays a critical role in the integrated stress response by transiently attenuating the translation of most mRNAs but promoting the translation of selected mRNAs, including those of transcription factors and genes related to nutrient metabolism, cellular adaptation, and autophagy. This regulation is indispensable for maintaining cellular homeostasis and responding to ER stress, highlighting the importance of EIF2S1 in canine health and disease.
