Porcine ganglioside-induced differentiation-associated protein 1 (GDAP1) is an integral membrane protein localized to the mitochondrial outer membrane and is implicated in the regulation of mitochondrial dynamics and cellular metabolism. GDAP1 plays a crucial role in maintaining the structure and function of mitochondria, which is essential for cellular energy production and cellular homeostasis. Mutations in GDAP1 have been associated with Charcot-Marie-Tooth disease (CMT), a hereditary peripheral neuropathy characterized by demyelinating and axonal forms, with GDAP1 mutations causing the most frequent recessively inherited demyelinating subtype CMT4A1. GDAP1 possesses two glutathione transferase (GST)-like domains and a C-terminal hydrophobic anchor that spans the outer mitochondrial membrane. Although its enzymatic activity as a GST remains controversial, GDAP1 is believed to bind GST substrates and influence cellular metabolism. The protein's function is tied to mitochondrial dynamics, where its overexpression leads to mitochondrial fragmentation, and its knockdown results in mitochondrial elongation. This regulation is dependent on the activity of dynamin-related protein 1 (DRP1), a key factor in mitochondrial fission.
