Porcine glycine amidinotransferase, also known as AGAT, is a critical enzyme involved in the biosynthesis of creatine, a key molecule in energy metabolism, particularly in skeletal muscle and the brain. This enzyme catalyzes the transfer of the amidino group from arginine to glycine, resulting in the formation of guanidinoacetate, which is then methylated to form creatine. Creatine is essential for the storage and transmission of phosphate-bound energy in muscle and brain tissues. In pigs, AGAT plays a vital role in maintaining energy homeostasis. Deficiencies in AGAT activity can lead to a range of metabolic disorders, including cerebral creatine deficiency syndromes (CCDS), which are characterized by severe neurological symptoms and developmental delays. The enzyme's function is not limited to creatine synthesis; it also impacts other metabolic pathways, such as the synthesis of homoarginine (HA), which has been linked to cardiovascular health.
