Porcine nicotinamide nucleotide transhydrogenase (NNT) is a mitochondrial enzyme that plays a critical role in maintaining cellular redox homeostasis and regulating metabolic pathways. NNT catalyzes the reversible transfer of hydride ions between NADH and NADP+, thus linking the mitochondrial protonmotive force to the generation of NADPH, an essential antioxidant and reductive power source for biosynthetic pathways. The forward reaction of NNT generates NADPH in the mitochondrial matrix, which is vital for combating oxidative stress and supporting anabolic processes. On the other hand, the reverse reaction, which oxidizes NADPH, also occurs under physiological and pathological conditions and is linked to the regulation of cellular metabolism.
