Canine carbamoyl phosphate synthetase 1 (CPS1) is a critical enzyme in the urea cycle, catalyzing the first and rate-limiting step where it converts ammonia and bicarbonate into carbamoyl phosphate. This reaction is essential for detoxifying ammonia in dogs, as it marks the entry point of nitrogen into the urea cycle, which is the primary pathway for nitrogen disposal. CPS1 requires the cofactor N-acetyl-L-glutamate (NAG) for activation, which is synthesized from acetyl-CoA and glutamate. Deficiencies in CPS1 can lead to hyperammonemia, a condition characterized by elevated blood ammonia levels, which can result in severe neurological symptoms and even death if untreated.
