Target Information
Canine sodium/temperature-inducible kinase 2 (SIK2) is a serine/threonine protein kinase belonging to the sucrose non-fermenting 1/AMP-activated protein kinase (SNF1/AMPK) family. It plays a significant role in various biological functions, including cell cycle regulation, tumorigenesis, and metabolism. SIK2 is highly expressed in multiple types of tumors, indicating its pivotal effect in oncogenesis, and it has been implicated in several signaling pathways such as the PI3K-Akt-mTOR pathway, the Hippo-YAP pathway, the LKB1-HDAC axis, and the cAMP-PKA axis. Recent studies have underscored its role in regulating cellular metabolism and survival, particularly in the context of cancer, where it couples metabolism with survival at the adipocyte-rich metastatic niche. SIK2 is also recognized as a potential therapeutic target for cancer treatment, with small-molecule inhibitors showing promise in rescuing the oncogenicity of SIK2 and reversing its abnormal activation of downstream pathways.
