Canine TANK binding kinase 1 (TBK1) is a serine/threonine kinase that plays a pivotal role in the regulation of cellular processes, including innate immunity, inflammatory cytokine production, autophagy, and cell survival/proliferation. TBK1 is a key component in the type I interferon response and is involved in the signaling pathways triggered by viral nucleic acid sensors such as RIG-I, cGAS, and TLR3. It is activated by pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs), and its activity is regulated by phosphorylation, ubiquitination, and SUMOylation. TBK1's role in the activation of transcription factors IRF3 and IRF7 is crucial for the production of type I interferons, which are essential in the antiviral immune response. In dogs, TBK1's function in immune modulation and its potential as a target for therapeutic intervention in diseases with disrupted immune responses, particularly viral infections, is of significant interest.
