Target Information

Porcine L-2-hydroxyglutarate dehydrogenase (L2HGDH) is a key enzyme involved in the mitochondrial metabolism, catalyzing the conversion of L-2-hydroxyglutarate (L-2-HG) into 2-oxoglutarate (2-OG). Disruptions in this enzymatic activity, such as those caused by mutations or reduced expression, can lead to the accumulation of L-2-HG, which is associated with neurometabolic disorders like L-2-hydroxyglutaric aciduria (L-2-HGA) and contributes to renal cell carcinoma (RCC) development. In the context of cancer, L-2-HG's ability to inhibit α-ketoglutarate-dependent dioxygenases impacts DNA and histone methylation patterns, thereby promoting oncogenic processes. The enzyme's role in renal cancer is significant, as increased L-2-HG levels are often a result of L2HGDH downregulation, positioning it as a potential therapeutic target. Furthermore, L-2-HG has been shown to alter amino acid metabolism in colorectal cancer through the mTOR-ATF4 pathway, affecting cellular metabolism and survival.
