Target Information

Porcine occludin is a vital protein component of tight junctions in the intestinal epithelium, playing a critical role in maintaining intestinal barrier function. It is a key determinant of the intestinal physical barrier, primarily composed of claudin, occludin, and junctional adhesion protein molecule-A (JAM-A), along with associated proteins such as zonula occludens (ZO)-1, Myosin, F-actin, and Myosin light chain kinase (MLCK). In pigs, Occludin's function is essential in attaching adjacent cells together, forming intercellular seals that prevent the entry of harmful substances into the circulation system while allowing the selective absorption of nutrients. It has been identified as a Porcine Epidemic Diarrhea Virus (PEDV) entry factor, highlighting its role in virus infection and pathogenesis. PEDV infection results in the disruption of tight junctional distribution of occludin, internalizing it from its position at the cell membrane to an intracellular location. Overexpression of occludin in target cells increases their susceptibility to PEDV infection, while the ablation of occludin expression significantly reduces their susceptibility, indicating its essential role in PEDV infection at postbinding stages.
