Target Information
Porcine serine hydroxymethyltransferase 1 (SHMT1) is a critical enzyme involved in one-carbon metabolism, catalyzing the reversible conversion of serine to glycine while producing 5,10-methylenetetrahydrofolate, a key molecule in the folate cycle. This enzyme plays a vital role in various biological processes, including DNA synthesis, repair, and methylation reactions. In pigs, SHMT1 is essential for primary-root growth, particularly under low-sucrose conditions. The mutation of SHMT1 leads to decreased sugar levels, excessive accumulation of reactive oxygen species (ROS), and severe root-growth arrest in the absence of sucrose. Conversely, overexpression of SHMT1 results in increased sugar levels, decreased ROS, and longer primary roots. Exogenous sucrose supply can fully restore root growth in SHMT1 mutants, suggesting that SHMT1 accumulation in roots is critical for sucrose accumulation and root growth.
