mRNA Display Platform Overview

The mRNA display is a tool for generating a diversity of protein and peptide libraries (ranging from thousands to trillions of unique sequences) for in vitro selection and direct evaluation. At BioVenic, we offer a specialized mRNA display platform for clients to discover therapeutic and novel proteins and peptides targeting various animal diseases. This method selects and evolves polypeptides with specialized functional properties through four main steps. At the end of this process, the library will be enriched with functional sequences. Cloning and sequencing will identify the selected functional proteins or peptides. mRNA display facilitates the discovery of new functional proteins or peptides without the need for prior design.

  • Transcription and Ligation
  • A double-stranded DNA library containing randomized nucleotide sequences is transcribed into an mRNA library, which is then ligated to a DNA fragment with puromycin attached to its 3' end.
  • Assist clients with the design and synthesis of customized double-stranded DNA libraries with randomized nucleotide sequences.
  • Provide quality control and validation of DNA library services to ensure sequence diversity and integrity.
  • Offer library design to optimize targeting properties and applications for specific animal diseases.
  • Transcription and Purification
  • The ligated mRNA Library is then translated in vitro. As ribosomes read RNA and not DNA, the ribosome stalls at the mRNA-DNA interface.
  • Supply optimized in vitro translation systems tailored to different mRNA libraries.
  • Monitor and analyze the efficiency and fidelity of translation in real time.
  • Troubleshoot and optimize translation conditions to maximize output and minimize stalling.
  • Selection (Affinity)
  • The fusion library undergoes a selection process, typically via co-immunoprecipitation with a target bound to beads, to remove proteins with low affinity for the target.
  • Develop and implement the co-immunoprecipitation systems using client-specified targets bound to beads.
  • Conduct high-throughput screening to identify high-affinity interactions.
  • Perform data analysis and report on selection efficiency, including providing recommendations for process refinement.
  • RT-PCR
  • Proteins that bind to the beads are reverse-transcribed into a new DNA library, which forms the basis for subsequent selection rounds.
  • Conduct reverse transcription of selected proteins back into DNA for generating new libraries.
  • Refine and enhance libraries iteratively, based on selection outcomes.

  • Each peptide or protein is physically linked to its encoding mRNA, allowing for immediate identification of the sequence following selection.
