Overview DNA Display Services Features Service Workflow Why Choose Us

DNA display is an in vitro selection system that identifies protein or peptide ligands from a vast library, with each ligand displayed alongside its encoding DNA. This technology facilitates rapid screening of a wide array of protein or peptide ligands for their corresponding receptors, overcoming the limitations of bacterial cells or RNA tags. BioVenic specializes in creating customized DNA libraries for the targeted screening of veterinary therapeutic proteins and peptides for various diseases. We offer a comprehensive one-stop service for discovering therapeutic proteins and peptides, meticulously tailored to meet the unique needs of our clients.

DNA Display Platform Overview

DNA display is an advanced cell-free technique in molecular biology for protein and peptide discovery, and it is increasingly used in the development of veterinary therapeutic drugs. It creates a library of DNA sequences, each encoding diverse proteins or peptides, which are lined to their corresponding DNA molecules. Thus, the direct link between the DNA and its encoded proteins or peptides is allowed to select the functional candidates. This method can overcome the technical limitations of conventional display technologies and has proved widely applied to screen diverse libraries of mutant protein and peptide ligands for various target receptors.

DNA Display Services

Description Services
  • DNA Library Construction
  • A DNA library encoding streptavidin-fused peptides is biotin-labeled and compartmentalized into water-in-oil emulsions along with an in vitro transcription/translation system.
  • Design and construct DNA libraries encoding target proteins or peptides.
  • Perform biotin labeling to facilitate subsequent steps and prepare materials for peptide synthesis.
  • The Conjunction of Protein/Peptide and DNA
  • Within each compartment, proteins or peptides fused to streptavidin are synthesized and linked to their template DNA via biotin.
  • Conduct in vitro transcription/translation reactions to synthesize peptides fused with streptavidin.
  • Ensure proper linkage of peptides or proteins to template DNA via biotin.
  • The Extraction of Conjunction
  • The DNA-protein/peptide conjugates are extracted from the emulsions.
  • Extract DNA–peptide/protein conjugates from water-in-oil emulsions.
  • Optimize extraction conditions to ensure high recovery and purification of DNA–peptide/protein complexes.
  • Affinity Selection
  • These conjugates undergo affinity selection against an immobilized bait.
  • Perform affinity selection assays using immobilized bait to identify peptides with specific binding capabilities.
  • Offer bait preparation and optimization services.
  • Washing and Elution
  • These conjugates are subjected to washing and elution steps to eliminate unbound molecules.
  • Provide optimized washing and elution protocols to efficiently remove non-binding molecules and recover bound DNA.
  • Perform post-elution DNA purification and concentration.
  • Multiple Rounds of Selection
  • The amplified DNA undergoes further rounds of selection.
  • Conduct additional rounds of selection to further enrich target molecules.
  • Selection Rounds or Sequencing
  • It is sequenced to identify successful ligands.
  • Perform high-throughput sequencing to identify successful ligands and their corresponding DNA sequences.
  • Offer data analysis and interpretation services.

DNA Display Platform Features

  • A vast library of DNA sequences encoding a variety of peptides or proteins is created.
  • The proteins or peptides are fused to streptavidin, facilitating their subsequent biotin labeling and selection processes.
  • Proteins or peptides are biotinylated and linked to their encoding DNA via biotin-streptavidin interaction, forming DNA–peptide conjugates.
