Canine polo like kinase 1 (PLK1) is a conserved mitotic serine-threonine protein kinase that plays a crucial role in the regulation of cell division, maintenance of genome stability, spindle assembly, mitosis, and DNA-damage response. Overexpression of PLK1 can lead to the undermining of cell cycle checkpoints, resulting in excessive cellular division and abnormal cell growth, which is a hallmark of cancer development. In dogs, PLK1 has emerged as a potential therapeutic target, particularly in tumors overexpressing c-Myc, where it coordinates cell proliferation, differentiation, self-renewal, and apoptosis. Inhibiting PLK1 expression through antibodies, RNA interference, or kinase inhibitors can reduce tumor cell proliferation and induce apoptosis in cancer cells without affecting healthy cells, making it an attractive target for drug development in veterinary oncology.
