Target Information

Porcine monoglyceride lipase (MGL), also known as porcine monoacylglycerol lipase, is a serine hydrolase that plays a pivotal role in the degradation of monoacylglycerols, particularly the hydrolysis of 2-arachidonoylglycerol (2-AG), an endocannabinoid. This enzyme is widely distributed in various cellular structures such as the plasma membrane, endoplasmic reticulum, and lipid droplets, and it significantly impacts lipid metabolism and the metabolism of polyunsaturated fatty acids. The inhibition of MGL has been shown to suppress the proliferation of certain cancer cells, highlighting its potential as a promising target for cancer treatment. The development of covalent small molecule inhibitors of MGL has progressed rapidly, offering a new approach to targeted cancer therapy with the advantages of strong covalent binding, high affinity, long duration of action, low dosage, and reduced risk of drug resistance.
