Target Information
Porcine retinoblastoma binding protein 9 (RBBP9) is a serine hydrolase that interacts with the retinoblastoma susceptibility protein (Rb). It is implicated in the development of pancreatic neoplasia and affects cell cycle control. RBBP9 is involved in the TGF-β signaling pathway, and its serine hydrolase activity is required for the development of pancreatic carcinomas. This activity inhibits TGF-β antiproliferative signaling through the suppression of Smad2/3 phosphorylation. Structurally, RBBP9 belongs to the α/β hydrolase superfamily of enzymes and has a crystal structure that confirms this fold, with a six-stranded parallel β-sheet flanked by α helixes. It contains a Ser75-His165-Asp138 catalytic triad, which is situated in a prominent pocket on the surface of the protein. The side chains of the LxCxE sequence motif, important for interaction with Rb, are mostly buried in the structure.
