Target Information

Canine 3-hydroxyisobutyryl-CoA hydratase (HIBCH) is a mitochondrial enzyme that plays a pivotal role in the catabolism of valine, an essential branched-chain amino acid. HIBCH catalyzes the hydrolysis of (S)-3-hydroxyisobutyryl-CoA (HIB-CoA), an intermediate in valine metabolism, which results in the production of the diffusible monocarboxylic acid (S)-3-hydroxyisobutyric acid from a CoA ester. This reaction is crucial as it protects cells from the toxic effects of methacrylyl-CoA, an intermediate immediately upstream of HIB-CoA in the valine pathway. HIBCH is not a member of the esterase family of enzymes and shows sequence similarity to the enoyl-CoA/isomerase family of enzymes, although it does not exhibit hydratase or isomerase activity. The activity of HIBCH is important for maintaining cellular redox balance and is implicated in the regulation of triacylglycerol accumulation in certain organisms. In dogs, HIBCH's function in valine metabolism is essential for energy production and detoxification processes, making it a significant target for understanding metabolic pathways and potential therapeutic interventions related to valine metabolism disorders.
