Target Information

Canine pantothenate kinase 2 (PANK2) is a vital enzyme involved in the initial step of coenzyme A (CoA) biosynthesis from pantothenic acid, also known as vitamin B5. This enzyme is particularly important in energy metabolism and is likely uniquely targeted to the mitochondria, which is significant for tissues with high-energy demands, such as the brain. In dogs, as in humans, mutations in PANK2 can lead to Pantothenate Kinase-Associated Neurodegeneration (PKAN), a progressive neurodegenerative disorder that affects movement, balance, speech, vision, cognition, and behavior. The disorder arises from debilitating mutations in the PANK2 gene and is characterized by a CoA deficiency that impacts critical neuronal processes, including iron metabolism and synaptic transmission.
