Veterinary Vaccine In Vitro Safety Assays

Inquiry Now

The in vitro safety assays of veterinary vaccines provided by BioVenic are aimed at assisting you in evaluating the safety of candidate veterinary vaccines at the cellular level. Typically conducted in the early stages of veterinary vaccine development, this assessment helps screen for potential safety issues, facilitating fine-tuning of vaccine formulations and production processes. BioVenic offers a variety of testing methods to assess the potential safety issues of vaccines to host cells.

Our Services

Cell Culture

BioVenic offers cell culture services for in vitro safety assays of veterinary vaccines. We have extensive experience in cell culture and employ advanced cell factory technology for large-scale cell cultivation. We provide well-characterized stable animal primary cell lines and offer cell line identification services.

  • Tailored to different specifications.
  • Microbe-free.
  • Low endotoxin levels.

Cytotoxicity Testing

Cell toxicity is a crucial parameter we employ to assess your veterinary vaccine's capacity to disrupt or kill cells. Measuring cell toxicity is of paramount importance in determining the impact of your veterinary vaccine on cell viability and ensuring safety post-administration. The methods provided by BioVenic for measuring cell toxicity not only directly assess cell toxicity but also indirectly detect it by measuring indicators such as the number of healthy cells within a cell population. Furthermore, we evaluate the integrity of cell membranes within the cell population, with a focus on detecting enzyme activity that leaks into the extracellular matrix when cell membranes are damaged.

Services Description
Cell Morphology Observation Using a microscope, evaluate the health status of cells by observing changes in cell morphology, such as shrinkage, expansion, or rupture.
LDH (Lactate Dehydrogenase) Assay Detecting LDH released into the culture supernatant when cells are damaged or ruptured.
SRB (Sulforhodamine B) Assay Utilizing the chemical binding of the Sulforhodamine B dye to proteins under mildly acidic conditions and subsequent extraction under alkaline conditions. The amount of extracted dye represents the number of cells in the sample, used to analyze the toxicity of the veterinary vaccine to cells.
MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) Assay Quantitatively measuring the reduction of MTT to a purple product by live cells when MTT is added to host cell cultures, allowing assessment of cell survival post-veterinary vaccine treatment.
Live Cell vs. Dead Cell Staining Combining various dyes, such as staining dead cells with ethidium homodimer and marking live cells with esterase-cleavable dyes.

Genotoxicity Testing

Genotoxicity refers to the toxic changes caused by veterinary vaccines to the genetic material (DNA, RNA) of cells, resulting in the breakage and disruption of nucleic acid chains. BioVenic offers genotoxicity testing services, conducting in vitro experiments to determine potential damage to genetic material.

Ames Test

The Ames test employs Salmonella typhimurium strains with mutations in histidine synthesis genes. These strains lack the ability to produce histidine and require it for growth. The test assesses the veterinary vaccine's potential to induce mutations that allow the cells to revert to a "prototrophic" state, enabling growth on a medium without histidine.

In Vitro Micronucleus Test

The in vitro micronucleus test is a genetic toxicity assay used to detect whether mammalian cells produce micronuclei after exposure to a test substance. BioVenic provides CHO cell lines or mouse lymphoma cells, exposing the target cells to the tested veterinary vaccine in a culture medium. The formation of micronuclei, additional small nuclei resulting from chromosome breakage, is observed in the cells.

Pro-inflammatory Cytokine Test

BioVenic offers in vitro detection services for pro-inflammatory cytokines in veterinary vaccines, including the assessment of the vaccine's impact on the pro-inflammatory immune response in an in vitro environment. The service involves cultivating specific immune cells or other relevant cells, such as macrophages and lymphocytes, exposing them to the tested veterinary vaccine, and subsequently evaluating the vaccine's influence on animal pro-inflammatory cytokines, such as IL-2, IL-6, IFN-γ, and TNF-α.
