Introduction of T7 Endonuclease 1 (T7E1) Assay

T7 endonuclease 1 mismatch assay is an efficient and cost-effective method for estimating the relative gene editing efficiency in cell populations, or for screening various clones and conditions. T7 endonuclease 1 is a phage-derived mismatch-specific enzyme that cleaves mismatched cruciform DNA, heteroduplex DNA, branched DNA, and Holliday junctions. Following a gene editing experiment, the genomic DNA around the target site is amplified by PCR, and the PCR amplicon is denatured and re-annealed by heating and slow cooling. If a Non-homologous end joining (NHEJ) event occurs, multiple products may be generated upon reannealing. Heteroduplexes are formed when the WT strand is reannealed to an indel-carrying strand that results in a mismatch. The heteroduplex product with mismatches is cleaved by T7 endonuclease. Separation of the DNA product by gel electrophoresis after treatment with T7 endonuclease will produce a band pattern indicative of the number of heteroduplexes in the sample. The amount of cleaved heteroduplexes is directly related to the amount of indel activity.
