Prime-editing Guide RNA (pegRNA) Design and Synthesis
The pegRNA consists of a fragment homologous to a genomic target sequence and a region to perform editing. It can be regarded as a specially modified sgRNA, whose sequence length and other designs not only affect the positioning of the editor, but also affect the editing efficiency. It has less stringent requirements for PAMs due to the varying lengths of RT templates, and it does not involve non-target editing.
